Background
"Sphinogosine Kinase has two isozymes (SPHK1 and SPHK2) that catalyze the phosphorylation of sphingosine into sphingosine1-phosphate (S1P). Sphingosine 1-phosphate mediates many cellular processes including migration, proliferation and apoptosis, and also plays a role in several types of cancer by promoting angiogenesis and tumorigenesis. SPHK is an important signaling enzyme which is activated by diverse agents, including growth factors that signal through receptor tyrosine kinase, agents activating G protein-coupled receptors, and immunoglobulin receptors. It also plays a role in breast cancer proliferation and chemoresistance. SPHK1 and SPHK2 are ubiquitously expressed. SphK-1 has been shown to mediate cell growth, prevention of apoptosis, and cellular transformation, and is upregulated in a variety of human tumors. In contrast, SphK-2 increases apoptosis, and may be responsible for phosphorylating and activating the immunosuppressive drug FTY720"
