Background
"Thymidylate synthase (TS) is a key enzyme in the de novo synthesis of 2′-deoxythymidine-5′-monophosphate (dTMP). TS catalyzes the methylation of 2′-deoxyuridine-5′-monophosphate (dUMP) to dTMP for which 5,10-methylene-tetrahydrofolate (CH2-THF) is the methyl donor.. TS plays a crucial role in the early stages of DNA biosynthesis. With inhibition of TS, an imbalance of deoxynucleotides and increased levels of dUMP arise. Both cause DNA damage Theenzyme has been of interest as a target for cancer chemotherapeutic agents. It is considered to be the primary site of action for 5-fluorouracil, 5-fluoro-2-prime-deoxyuridine, and some folate analogs. Expression of this gene and that of a naturally occurring antisense transcript, mitochondrial enolase superfamily member 1, vary inversely when cell-growth progresses from late-log to plateau phase. Polymorphisms in this gene may be associated with etiology of neoplasia, including breast cancer, and response to chemotherapy. TS presents a novel biomarker for diagnosis and prognosis in patients with pancreatic cancer."
